A new alternative to produce gibberellic acid by solid state fermentation BABT

Gibberellic acid (GA3) is an important hormone, which controls plant's growth and development. Solid State Fermentation (SSF) allows the use of agro-industrial residues reducing the production costs. The screening of strains (four of Gibberella fujikuoroi and one of Fusarium moniliforme) and substrates (citric pulp, soy bran, sugarcane bagasse, soy husk, cassava bagasse and coffee husk) and inoculum preparation study were conducted in order to evaluate the best conditions to produce GA3 by SSF. Fermentation assays were carried out in erlenmeyers flasks at 29°C, with initial moisture of 75-80%. A simplified model for A. Niger FS3 growth during phytase formation in solid State fermentation BABT

A simplified model to describe fungal growth during citric pulp fermentation for phytase production was described for the first time. Experimental data for biomass growth were adjusted to classical mathematical growth models (Monod and Logistic). The Monod model predictions showed good agreement with the experimental results for biomass concentration during 96 hours of fermentation. Parameters such as yield of biomass from oxygen (Y X/O), maintenance coefficient (m) and specific growth rate (µ) were compared showing a good correlation between the data and the model. An alternative method for biomass determination in this process was developed since a great correlation was found between biomass growth and enzyme formation.

Acid and enzymatic hydrolysis to recover reducing sugars from cassava bagasse: an economic study BABT

The objective of this work was to study the acid and enzymatic hydrolysis of cassava bagasse for the recovery of reducing sugars and to establish the operational costs. A statistical program "Statistica", based on the surface response was used to optimize the recovery of reducing sugars in both the processes. The process economics was determined considering the values of reducing sugars obtained at laboratory scale, and the operations costs of a cylindrical reactor of 1500 L, with flat walls at the top and bottom. The reactor was operated with 150 kg of cassava bagasse and 1350 kg of water. The yield of the acid hydrolysis was 62.4 g of reducing sugars from 100 g of cassava bagasse containing 66% starch. Alpha amylase from a fungal culture grown on oil cakes and its properties BABT

Solid-state fermentation was carried out for the production of alpha-amylase using Aspergillus oryzae. Different oil cakes such as coconut oil cake (COC) sesame oil cake (SOC), groundnut oil cake (GOC), palm kernel cake (PKC) and olive oil cake (OOC) were screened to be used as substrate for the enzyme production and also compared with wheat bran (WB). GOC was found to be the best producer of the enzyme among these. Combination of WB and GOC (1:1) resulted higher enzyme titres than the individual substrates. Maximum amount of enzyme (9196 U/gds) was obtained when SSF was carried out using WB + GOC, having initial moisture of 64% and supplemented with lactose and ammonium nitrate (1% each) at 30ºC for 72h using 2 mL spore suspension (6x10(7)spores/ml)....

The purpose of this work was to reduce the cost and improve the quality of the plant material during the micropropagation process in tissue culture. Partially improvement in the rooting process, coupled with cost reduction was obtained during the invitro rooting by the use of a natural support based on sugarcane bagasse as a substitute for the traditionally used agar gelled medium. Azospirillum sp . inoculation in wheat, barley and oats seeds greenhouse experiments BABT

The aim of the present work was to evaluate the inoculation effects of Azospirillum sp. RAM-7 strain, in seeds of wheat, barley and oats cultures on yield and total nitrogen content of the grains in greenhouse experiments. Seeds inoculation with Azospirillum sp. RAM-7 strain enhanced grain yields, even so, the responses vary among the evaluated cultures. For wheat, significant increases were obtained when the inoculation was associated to 100% of the recommended nitrogen, although the treatment that received only inoculation produced 7.4% above the control, however this difference wasn't significant. Biofiltration of a styrene/acetone vapor mixture in two reactor types under conditions of styrene overloading BABT

This aim of study was to compare the performance of a biofilter (BF) and trickle bed reactor (TBR) under increased styrene loading with a constant acetone load, 2 gc/m3/h. At styrene loading rates up to 30 gc/m3/h, the BF showed higher styrene removal than TBR. However, the BF efficiency started to drop beyond this threshold loading and could never reach steady state, whereas the TBR continued to yield a 50% styrene removal. The acetone removal remained constant (93-98%) in both the reactors at any styrene loading. Once the overloading was lifted, the BF recovered within 26 min, whereas the TBR efficiency bounced back only to 95%, gradually returning to complete removal only in 10 h.

Bovine mastitis in the metropolitan area of Curitiba: antibiotic resistance and antimicrobial control of the infection BABT

A study from cows with mastitis was performed and Staphylococcus aureus was the predominant pathogen in 46.4 % among 153 studied strains from 276 milk samples of infected cows. Antibiotic resistance of 71 S. aureus isolates was determined in order to search resistant strains to antibiotics of clinical interest, as well as to determine their degree of multi-resistance. It was found that 60% of the S. aureus strains presented resistance to β-lactams, but none to oxacillin, teicoplamin or vancomycin. On the other hand, with the aim of reducing the use of current antibiotics and their associated resistance, a new formulation was introduced. Coffee residues as substrates for aroma production by Ceratocystis fimbriata in solid state fermentation BJM

The ability of two different strains of Ceratocystis fimbriata for fruity aroma production by solid state fermentation (SSF) was tested on coffee pulp and coffee husk complemented with glucose as substrates. Experiments were carried out in 250 mL Erlenmeyer flasks and the experimental conditions were: 70% of initial moisture, 20% of glucose addition and pH 6.0. Aeration was made by passive diffusion through the gauze covering the flasks. Headspace analysis of the culture by gas chromatography (GC) showed that 12 compounds were produced with coffee husk. ABSTRACT Nidularium procerum and Nidularium innocentii (Bromeliaceae) were cultivated in vitro on media supplemented with different sources and levels of GA3 (gibberellic acid). These sources were the commercial powder (analytical degree) and fermented extract obtained by Fusarium moniliforme via solid state fermentation. The in vitro elongation and rooting of these plants were evaluated after 50 days of cultivation. The GA3 present in the fermented extract possess the same effect of purified GA3 (analytical degree) for the increase of the height of aerial part of shoots of N. innocentii, but not for the N. procerum being the GA3 fermented extract in a lesser degree. Aedes aegypti is the vector of Dengue disease, responsible for 20,000 deaths/year worldwide. Bacillus thuringiensis var israelensis - Bti releases selective and effective toxins (crystal proteins) against A. aegypti larvae. We present a low cost bioprocess for toxin production, accomplished by a selected Brazilian strain Bti (BR-LPB01) and employment of low cost substrates. Soybean meal and sugarcane molasses lead to high toxic effectiveness after 2L bioreactor fermentation (LD50=26ng/mL), near to the reference strain IPS82 (LD50=17.3 ng/mL). Both the raw material costs and purification steps were minimized. Firstly, medium composition for the growth of Corynebacterium glutamicum ATCC 21799 was optimized targeting enhanced L-Lysine production - salt, vitamins and nitrogen sources concentrations were tested and selected. Effect of caffeine and tannins on cultivation and fructification of Pleurotus on coffee husks BJM

The objective of this work was to study the effect of caffeine and tannins on Pleurotus sp. cultivation and to evaluate the feasibility of using coffee husks as substrate for mushroom cultivation. Eight strains of P. ostreatus and two strains of P. sajor-caju were screened on a medium prepared from agar extract of coffee husk. Based on best mycelial growth and biomass production, the strain P. ostreatus LPB 09 was selected for detailed studies. With the increase of caffeine concentration, the mycelial growth and the biomass production decreased, and no growth was observed when concentration of caffeine was 2500 mg/L.
